Endotoxin Testing — Why It's Non-Negotiable
Bacterial endotoxin can distort biological results at trace levels even when mass and chromatographic-purity results look acceptable.
Endotoxins are lipopolysaccharides from the outer membrane of Gram-negative bacteria. They are shed when bacteria grow and die, can enter a process through water, raw materials, or handling, and are potent immunostimulants at very low concentrations.
Trace amounts can distort biology
Controlled work shows that low endotoxin levels can drive immune-cell proliferation and expansion, producing false-positive signals in immunogenicity and inflammation assays. Recombinant-protein research contains repeated examples where an apparent activity was later traced to contaminating endotoxin rather than the protein under study.
The effect can outlast one assay
Endotoxin is a classic immunological adjuvant. Beyond an acute inflammatory response, it can contribute to an adaptive, antigen-directed response. Anti-drug antibodies can lower effective exposure and inhibit biological activity, so an earlier sensitizing exposure can complicate later work with a cleaner lot.
Identity and purity do not answer this question. Endotoxin requires its own assay, including suitable interference controls for the tested matrix. It is the third part of the initial Radix analytical panel because 'the right molecule' and 'chromatographically clean' are only two of the questions that matter.
